In February, I saw Michael Browning present the results of PAX-D, a randomised controlled trial of pramipexole in treatment-resistant depression Like many in the audience, I was blown away by the results (link in bio)

Systematic review and meta-analysis: Stopping antidepressants leads to slight symptom increases compared to placebo, but these symptoms are not clinically significant and do not include mood changes. ja.ma/4lNdBIu

Playing Whack-a-Mole With the Uncertainties of Antidepressant Withdrawal Complacency is the wrong conclusion of a new debated study psychiatrymargins.com/p/playing-whac…

“Basically, if you are still someone who treats the Davies and Read estimate of antidepressant withdrawal as valid—take the L and move on! The data from clinical trials just doesn’t back it up.”

Big late stage win for Takeda! Oveporexton (TAK-861) hits on all endpoints in narcolepsy for the first-in-class orexin receptor 2 selective agonist. Congrats to Andy Plump and the team at $TAK - perseverance and conviction on the MoA paid off takeda.com/newsroom/newsr…

Can administrative real-world data substitute for RCTs and offer propensity matching to reduce bias? Results often fail to correspond & Lars G. Hemkens et al study showed direction of effect differed in 31% & CI failed to include RCT estimate in 56% pubmed.ncbi.nlm.nih.gov/26858277/

Pramipexole vs. quetiapine vs. amantadine in TRD. Interesting open label, short term RCT using a surprisingly low dose (I suspect that the reported dose of 37.5mg of pramipexole is a typo and it should be 0.375mg) sciencedirect.com/science/articl…

1/ Just out in Nature Medicine : we show the opioid system modulates ketamine’s effects in major depressive disorder, with naltrexone pre‑treatment reducing acute glutamatergic activity and acute antidepressant effects. nature.com/articles/s4159…

New preprint alert with Michael Browning and Chamith Halahakoon! People have been using online RL tasks with affect probes. But to what extent do they reflect real-life affective experience? We tested this with 3 weeks of EMA + RL tasks in 339 participants.

Taking part in research is good! Well done to the PAX-D team and of course to the participants who make clinical research possible I think the rate-limiting step is not the willingness of patients to participate but unnecessary bureaucratic hurdles we place before them
