Li Li Fernando Camargo Allon Klein Lab Cell We believe that this is the first direct demonstration of strong epigenomic memory at single-cell level with lineage tracing. DARLIN and Camellia-seq could open a lot of possibilities for studying cell lineages in different context.
Li Li Fernando Camargo Allon Klein Lab Cell To our surprise, we found that the transcriptomic memory fades away quickly over time (so does the memory with chromatin accessibility), while the memory associated with DNA methylation persists even after 2 months, despite major changes of celluar environment in this process!

Li Li Fernando Camargo Allon Klein Lab Cell If the memory is strong in a given modality (say transcriptome), then the progenies of two different clone would be clearly separated in the corresponding embedding. Otherwise, these progenies would be mixed.
Li Li Fernando Camargo Allon Klein Lab Cell Finally, we asked a very interesting question about memory or inheritence: will cells from the same HSC clone have similar transcriptomic or epigenomic profiles? Or will the progenies of a cell 'remember' that they come from the same mother? We test this systematically with HSCs

This is a fabulous collaboration between Allon Klein Lab and shouwen wang, Li Li, and Fernando Camargo. The DARLIN mice is superior to CARLIN, and the single-cell multi-omic lineage tracing method is a great addition to single-cell genomics toolkits!
