We found the activity of GeoCas9 is highly dependent on the availability of free Mg2+. In vitro at high [Mg2+](5mM), GeoCas9 and iGeoCas9 activity were similar. At low [Mg2+](0.1 mM), iGeoCas9 sustains activity while GeoCas9 R-loop formation rate drops to undetectable levels. 3/7
