Mfn2 tethers ER to mitochondria, but what's its partner on the ER? How does it tether heterotypic organelles without fusing them? Deborah discovered that Mfn2 is alternatively spliced in two variants Ermin2 and Ermit2 that lack the GTPase domain and go only to the ER. 2/

Ermin2 and Ermit2 do not influence mitochondrial morphology. Ermin2 regulates ER shape and luminal connectivity. Ermit2 is enriched in MAMs. Ermit2 increases ER-mito proximity measured by ddGFP, FEMP, EM. In cells, Ermit2 supports Ca2+ transfer from ER to mitochondria 3/

In the liver, Ermit2 facilitates ER-mitochondria phospholipid transfer, reduces ER stress caused by Mfn2 ablation and in models of NASH. Molecularly, Ermit2 CC2 domain is required to interact with CC1 and CC2 of mitochondrial Mfn1&2, in a complex apt to stabilize ER-mito 4/
