Has anyone got experience with affinity tags that work at low pH (around 5)?

Does anyone know why in ion exchange protocols you are supposed to first run some 100% B over the column and then equlibrate with 0% B before sample loading? What's the point of that?

A bit of random SciArt from my last LCP crystallisation screen. I call this one "One eyed LCP moon" which also involved sacrificing some blood during the making of this work.

Okay this kept me up all night: I left a peristaltic pump running overnight for binding on an affinity column at 1 ml/min, assume no dead volume/CV and 100% binding in 400 ml of solution. If it's on a cycling motion how long does it take until only 10% of protein are in solution?

Always fun purifying a GFP tagged protein!
