Probing the haem compartment further, we detail haematopoietic stem progenitor cell (HSPC) emergence from hemogenic endothelium, also observed in public yolk sac iPSC scRNA-seq (Clara Alsinet Armengol et al rdcu.be/cTfOt) similar to aorto-gonado-mesenephros (AGM) [3/9]
Our data are consistent with definitive HSPC predominance over primitive by 8 PCW (Vincenzo Calvanese et al rdcu.be/cTgFF) We find differential linage potentials before and after AGM contributions to haematopoiesis in YS [4/9]

We find distinct waves of HSPC (red👇) lineage potential for different blood/immune cells before/after AGM contributions to haematopoiesis in YS. Our data are consistent with definitive HSPC predominance over primitive by 8 post conception weeks (Vincenzo Calvanese et al) [4/12]
We observe a pre-AGM accelerated pathway to macrophage differentiation in the YS prior to monocyte emergence (also in iPSC scRNA-seq Clara Alsinet Armengol et al) Macrophages in the YS post-AGM are from proliferating macrophage pool and monocyte differentiation [5/12]
