ST-EPNs segregate into molecularly and clinically distinct subgroups, hinting at different developmental roots. But what are the developmental signatures and microenvironmental factors driving these tumors’ behaviors? 2/

To answer these questions, we integrated single-cell & spatial transcriptomics with live cell imaging to map cell states, spatial organization, and dynamic behavior. 3/

We found that tumor subgroups harbor two distinct progenitor-like cell states reminiscent of early human brain development and diverge in the extent of neuronal vs ependymal differentiation. 4/

Next, we used 10X Xenium to characterize where these cells are, and by combining with H&E we could derive transcriptionally and morphologically distinct regions such as these distinct ependymal rosettes: 5/

Similar to findings from Tirosh Lab in gliomas, we see distinct structured vs non-structured regions that are driven by MES/hypoxia, with recurrent patients having more structure and more MES! 6/

Check out this exciting project deciphering cellular and spatial heterogeneity and neuronal influence on different cell states in supratentorial ependymoma! Biggest congrats to everyone involved!

Couldn’t be happier to see our work on resolving cellular and spatial heterogeneity, and the influence of the microenvironment on driving cell states in pediatric ependymomas out as a preprint 🔥 biggest congrats to Daeun Jeong, Mariella Filbin and all the collaborators 🥳

Thrilled to share our work Mariella Filbin, @GojoJohannes, and Carl Koschmann, now published in Cancer Cell. We present a translational approach identifying the PDGFRA inhibitor avapritinib as a novel targeted treatment for pHGG with PDGFRA alterations. cell.com/cancer-cell/fu…
