We recently discovered our favourite fast growing strain New England Biolabs Turbo (RecA+) creates a mess of concatemers, using plasmidsaurus. These disappear in RecA- strains (DH5ɑ, Top10 etc.) but they are all so slow! Suggestions for a fast growing RecA- strain? Addgene Tom Ellis

My plasmid was giving unexpected restriction patterns. Thanks to plasmidsaurus, I realized there was a bacterial insertion between the KanR gene and the replication origin. Apparently, this is a common phenomenon in some plasmids (eg. pCambia-based)🤯doi.org/10.1111/1751-7…
