We studied the neuronal anatomy of planarian with single cell precision using tissue expansion and TLSM. We explored the possibility to illustrate the neural networks and regeneration using the latest tissue imaging techniques. Results are promising, but labeling is critical.

I think we understand tissue clearing and imaging much more than ~10 years ago. We can obtain results like below routinely. Making the tool more accessible and efficient, developing methods to use tissue structure data more efficiently appear to be major challenges in the next.

I propose a method to increase the imaging efficiency of tiling light sheet microscopy using scanning non-coaxial beam arrays synchronized with regional virtual confocal slits. It makes TLSM as fast as conventional LSM and keeps all rest advantages, but needs a new sCMOS camera.

There is a patented technology, invented by Bi-chang and me, to improve the imaging ability of lattice light sheet microscopy for large samples using tiling LLS . It can be conducted on any original design LLS microscope without hardware change. patents.google.com/patent/US20220…
