Mutational signature is an essential part of genomics, but small differences in analysis can make a big difference in conclusion. Friends at Peter Park lab Hu Jin and Doga Gulhan tackled several key issues: “flat” signatures, assignment, and validation in their new tool!

Beyond excited to see this paper out. Most satisfying paper reproducing lung adeno to small-cell transformation in experimental model. Key players: EGFR, TP53, RB1, PIK3CA/PTEN, and MYC. Huge congratulations to Eric Gardner, Ashley Laughney, and the team! science.org/doi/10.1126/sc…
Functional proof of one-step oncogenesis by BRD4-NUT in mice. 100% penetrance from squam epithelium. Cancer types driven by a single massive driver could melt down by effective targeted therapy. Hope one day we see differentiation syndrome in NUT! Cancer Research aacrjournals.org/cancerres/arti…
Very insightful story regarding the sex difference in Xp11 translocation RCC, more common in female. Hibernating Xp fragment where the TFE3 gene resides is waken up by the translocation to autosomes. Sex chroms are rich source of cancer biology. Congrats to Srinivas Viswanathan et al.!
