love this story from Denny May & Sangwon Yun - they develop a tool to define single-cell chromatin compaction states AND track chromatin remodeling in the same cells over hours/days. Here, differentiating stem cells in live skin! There's also imaging of transcriptional dynamics🤩

Super excited to be hosting Dr. Jennifer Raff at Yale Department of Genetics Yale School of Medicine tomorrow. I don’t think I’ve ever had a three hour dinner (along with the amazing Massilani Lab) feel like it went by in 20 min. Join us tomorrow in the Medical Historical Library at 4:15!

Excited to share our paper just published in Journal of Cell Biology! We find that cell cycle controls long-range Ca2+ signaling in the epidermal stem cell layer and this signaling feeds back into cell cycle progression. bit.ly/3oP6cjD 1/14

Mutations in two autism-linked genes with different functions cause similar effects on dopamine and neuroimmune pathways in the brains of zebrafish. This convergence could light the way for precision treatments in the future. By peter hess. bit.ly/3Mj2pmP

We’re pleased to share our first publication from the Sozen lab, now out nature! Through our 3D strategy, we capture spontaneous emb. + exembr. lineage bifurcation to study aspects of early human embryo patterning, starting from a single stem cell type. nature.com/articles/s4158…

The ‘human extra-embryoids’ (hEEs) consist of epiblast- & hypoblast-like cell types w/o trophoblast derivatives. We leverage this reductionalist system to dissect the signaling dynamics + early patterning events that are key to early human development. 2/n

Monique Pedroza (@pedroza_monique) 's Twitter Profile Photo

The hEE sees an initial segregation of epi- & hypo-like compartments (~78% efficiency). The hypo-like compartment plays an integral role in epi-like morphogenesis and we find that TGFβ/NODAL & FGF activity crucial for hypo-like specification. 4/n

This heterogeneity in expression patterning allowed us to probe BMP restrictive + inductive activity in the context of symmetry breaking in our system and explore the relationship between a few key players. 6/n

Monique Pedroza (@pedroza_monique) 's Twitter Profile Photo

Expectedly, BMP agonists most readily tipped the balance towards an amnion-like fate. Unexpectedly, we describe a novel role for anterior visceral endoderm-like population in amnion-like patterning that offers a new area of exploration. 7/n

Technologies to model early mammalian embryo development in vitro have advanced rapidly, providing new insights into self-organization and environmental control. Berna Sozen and I wrote a review as part of a special issue on #earlyembryonicmodels: rb.gy/fkpjk (1/3)
