3 and a half years ago, we reported that a partial loss of function VCP mutation results in buildup of tau aggregates causing dementia. Now we have identified small molecule VCP activators that help clear ubiquitinated protein aggregates. doi.org/10.1172/jci169…

Delighted to share our latest preprint where we describe first-in-class DUB #MolecularGlues by Francesca Chandler et al. Another amazing collaboration with Roger Greenberg Professor Francesco Del Galdo and Jo Salvino labs. biorxiv.org/content/10.110…

We screened for DUB inhibitors and identified a hit which did not reproduce -- this turned out to be a 'dirty' well in the library. Instead of dismissing it, we characterised it fully and discovered 1) was a highly selective inhibitor for JAMM-domain DUBs and 2) a molecular glue

Amazing efforts from the first author Francesca Chandler showed the mode of action by #cryoEM where 2 small molecules glue a 16-subunit protein complex. Animation credit Jeroen Claus Phospho
