Very impressed by Sébastien Levesque et al. for venturing into the CAR-T cells universe and generating such a solid set of data. Find something you like ⬇️ (i) saturation mutagenesis via prime-editing, (ii) modified Guide-Seq pipeline, (iii) xenograft model for CART-cell toxicity.

Thanks to Doris Duke Foundation for their visionary and unparalleled commitment to sickle cell research. Exciting times, but much left to do.

My thesis advisor at Brown University, Dr Susan Gerbi, my postdoctoral advisor, Dr Alan Wolffe, and my colleagues at Sangamo are the ones who should get this award. I will accept it on their behalf - eternally grateful for my good fortune. graduateschool.brown.edu/news/2024-05-1…

In this Brief Communication you can find: I) An easy way to boost PE efficiency in HSCs. II) A ddPCR assay for RT processivity. III) An optimized IVT template for PE mRNA. As we say in Italy: "good wine is kept in small barrels". Thanks to Sébastien Levesque & Daniel Bauer.

Excited to share our latest work, motivated by the challenge of efficient prime editing in HSCs. We find that the metabolic state of quiescent cells presents a hurdle—but it’s surmountable, by boosting dNTPs. Led by the incomparable Sébastien Levesque. rdcu.be/dJdgK

For 25 years, the Toll-like Receptor pathway was considered a series of distinct protein complexes that drive inflammation. Today, we report that the entire pathway, from receptor to transcription factor, is executed from within one complex—the myddosome. nature.com/articles/s4158…

Great collaboration between the Hughes Group and Watkins labs, reporting the first Gain of Function de novo enhancer causing Mendelian disease (ST Depression Syndrome), defining a multidisciplinary path with ML to solving non-coding clinical genetics. medrxiv.org/content/10.110…

First pre-print from the lab! Together with Luca Pinello and Daniel Bauer we developed CRISPR-CLEAR, an experimental and computational pipeline to dissect enhancers at nucleotide resolution. biorxiv.org/content/10.110…

1/9 Thrilled to share our latest work on CRISPR-CLEAR with Davide Seruggia and Daniel Bauer labs. Over the past years we've been improving CRISPR tiling screen resolution and we have something to show you! Check also out David's excellent tweetorial!

Delighted to share our #NightScience tale: "A tetramer of BCL11A is required for stable protein production & fetal hemoglobin silencing," in Science Magazine. Co-led by Zheng Ge, Yin Maolu, & me, here’s a tweetorial about our puzzle-solving adventure! 1/7 tinyurl.com/2tpaaua3
