We looked into the proteomics data and did not find a difference in Pol II concentration between the two species, though it's unclear how comparable the data is. For second question, we also observed low Pol II footprint in differentiated cell types.

Cell Reports Check out our existing paper, just published in Cell Reports showing that partially assembled TFIID complexes (without TAF7 or TAF10) can sustain Pol II transcription initiation but cannot replace holo-TFIID over several cell divisions and/or development.

how cool is that? it still amazes me how Victor decided in 1994 (!) to look at such small RNAs in the lin-4 mutant worms . and the complementarity of the two lab's discoveries (one contributing the small RNA, the other one the target sites) is just lovely. warmest gratulations!
