A really cool part of this work was understanding the effects of the intramolecular H-bonds on molecule conformation and therefore biological activity. In previous work (onlinelibrary.wiley.com/doi/full/10.10…) we showed that the intermolecular H- bond was beneficial for bromodomain binding.
