I am working with an IRES construct for the first time. I transformed in DH5a yesterday and after 24h the colonies have barely grown. Does anyone know if this is standard for IRES? In comparison, I have a 2A construct that has pickable colonies.

Had a guest stay the night. All before 7 AM while I was asleep, they managed to: shower and use my personal towels, use my loofah, douse themselves in my perfumes, steal my socks, steal my shoes, steal my underwear. Couldn’t get them out the door fast enough.
