🚀 I am thrilled to add another piece to the “ONECUT1 diabetes” puzzle, again with joint forces between the #JulierLab at #INSERM and the collaborative efforts #IvanCosta, the Institutes of #HumanGenetics and #TransfusionMedicine at #UlmUniversity, and other collaborators!

Our journey began with characterizing the regulatory patterns of ONECUT1 (OC1) during pancreatic lineage commitment, published in #CommunicationsBiology (doi.org/10.1038/s42003…).

We then discovered that OC1 coding mutations are associated with various forms of diabetes as modeled via pluripotent stem cell differentiation, details in #NatureMedicine (doi.org/10.1038/s41591…).

Building on this foundation, we're excited to announce our latest publication in #CellReports! Brilliant #SarahMerz completed her PhD with her second first authorship under the guidance of #SandraHeller, alongside #CecileJulier and me (doi.org/10.1016/j.celr…).

Here, we identified a disease-causing deletion upstream of OC1 in compound heterozygosity with an OC1 mutation in an affected individual, while investigating its functional role as a cis-acting enhancer influencing OC1 expression during pancreatic development.

We also explored in vitro therapeutic options for affected patients and linked a low-frequency variant to type 2 diabetes. Excitingly, the #DanweiHuangfuLab utilized CRISPRi screening during pancreatic differentiation, confirming our findings regarding the same enhancer.Congrats!
