We had previously shown that TBX3 and #WNT cooperate in limb #DevBio We now have a shocking finding 😲 TBX3 physically engages with β-catenin to drive metastasis genes in #ColorectalCancer (CRC) Look where TBX3 is expressed in hCRC > together with AXIN2 and in LGR5+ cells

We used CUT&RUN-LoV-U ♥️ ICEBERG 🧊 to map all TBX3 binding sites in CRC Many are the proto-oncogenes that seem to be regulated by TBX3!

And surprise 🫢 TBX3 and β-catenin binding patterns largely overlap! Look for example this poor oncogenesis-driver MYC ⬇️

We combined CAGE-seq, CUT&RUN and HiChIP-H3K27ac in CRC cells to identify the transcriptional instances directly regulated by TBX3/β-catenin Most targets are genes known to regulate metastasis in human CRC!!! 🫨🤯

Proximity proteomics shows that TBX3 becomes physically proximal to members of the #WNT transcriptional complex, including TCF7, TLE2, ARID1B and SMARCE1 (SWI/SNF - BAF)

How does TBX3 physically connect the #WNT complex? Structure inspection and #AlphaFold prediction uncovered a short Asp-Pro-Phe (NPF) motif Below the NPF within the crystal structure of TBX3-T-box Next to it the predicted CTHULHU👾monster shows how TBX3 might manage the complex
