Phase one of iGEM Toronto Wet Lab includes literature review and protein database analysis on PETase and other molecular components of its enzymatic pathway, as well as other candidate plastic-degrading enzymes. Via biochemical simulations, mutations for PETase will be developed.

Phase two : With their variants in hand, the Wet Lab aims to develop circuits and constructs that can express the proteins at the desired efficacy. These circuits and constructs will then be incorporated into an E.Coli chassis for further use in a bioreactor setting! #synbio

Phase 2 : “To validate the Wet Lab’s designs, the Dry Lab will be modelling the variants while remaining faithful to the natural protein environment. Ultimately, this will ensure whether the variants are viable for further use in large-scale, industrial-based situations.”

Wet Lab’s final stage includes designing a cost-effective bioreactor that can maximize the purified product yield as well as support the optimal efficacy of microbial degradation. This bioreactor can be used in industrial circular economies to degrade and obtain PET byproducts.
